Why would car tires have a knot on it?
There are steel cords inside the rubber. If one of these cords frays or breaks, it makes a bubble, or bump or all kinds of different stuff. REPLACE YOUR TIRE IMMEDIATELY. It's going to blow soon and you don't want to be stuck on the side of the road.

Occasionally these knots develop because of a manufacturing defect, in this case it will be covered under warranty.
